After training on the analyzers, feel free to sign up for time on the equipment (analyzers available for use at anytime for GT users, sorters available during peak hours: 9am-6pm). BD Fusion (Located in EBB):
The BD FacsAria Fusion is an air-cooled, multi-laser, sorter with the ability to acquire up to 17 fluorescent parameters at once. It houses 5 lasers and has a similar configuration as our BD Fortessa. Translating an experiment from the Fortessa to the Fusion is therefore simplified. The configuration includes the UV (355nm, 65 mW), violet (405nm, 100 mW), blue (488nm, 100 mW), yellow-green (561nm, 50 mW), & red (640nm, mW) lasers. The Fusion has capability to perform 2 way, 4 way, and plate sorting. The index sorting option is available for single cell plate sorts. This instrument can also be utilized for analysis only.
BD FacsAria IIIu (Located in Petit Institute):
The BD FACS Aria IIIu can analyze and sort cells and other particles between a range of 2-30 microns in diameter. It houses 4 lasers, and the laser configuration includes violet (405nm), blue (488nm), yellow-green (561nm), and red (640nm) lasers. The FacsAria IIIu has capability to perform 2 way, 4 way, and plate sorting. The index sorting option is available for single cell plate sorts. This instrument can also be utilized for analysis only.
Amnis Imaging Flow Cytometer (Located in Petit Institute Biotech Building):
The ImageStreamX Mk II System is a benchtop, multispectral, imaging flow cytometer designed for the acquisition of up to 12 channels of cellular imagery. By collecting large numbers of digital images per sample and providing a numerical representation of image-based features, the ImageStreamX Mk II combines the per cell information content provided by standard microscopy with the statistical significance afforded by large sample sizes common to standard flow cytometry. With the ImageStreamX Mk II System, fluorescence intensity measurements are acquired as with a conventional flow cytometer; however, the best applications for the ImageStreamX Mk II take advantage of the system’s imaging abilities to locate and quantitate the distribution of signals on in or between cells.
BD LSR II (Located in Petit Institute Biotech Building):
The BD LSR II is an air-cooled, multi-laser, benchtop flow cytometer with the ability to acquire up to 14 fluorescent parameters at once. It houses 5 lasers, which include the UV (355nm, 65 mW), blue- violet (445nm, 100 mW), blue (488nm, 100 mW), yellow-green (561nm, 50 mW), & red (640nm, mW) lasers. It uses a fixed-alignment system that transmits light through a flow cell to configurable octagon and trigon detector arrays.
BD Fortessa (Located in EBB):
The BD LSRFortessa is also an air-cooled, multi-laser, benchtop flow cytometer with the ability to acquire up to 17 fluorescent parameters at once. It houses 5 lasers, which include the UV (355nm, 65 mW), violet (405nm, 100 mW), blue (488nm, 100 mW), yellow-green (561nm, 50 mW), & red (640nm, mW) lasers. Similar to the LSRII, it uses a fixed-alignment system that transmits light through a flow cell to configurable octagon and trigon detector arrays. The High Throughput System (HTS) option for 96 well plates is available for use with this system.
BD Accuri C6 (Available in both IBB & EBB):
This 4 color, benchtop system is equipped with blue & red lasers, two light scatter detectors, & four fluorescence detectors with optical filters optimized for the detection of FITC, PE, PerCP, and APC. A compact optical design, fixed alignment, and pre- optimized detector settings make the system user-friendly. Voltage modification & optimization is not possible on this instrument. Particle size capability includes a range from 1um to 40um, with flow rates ranging from 14ul/min to 66ul/min.
LI-COR Odyssey CLx Far Infrared Imager (Available in both Petit Institute Biotech Building & EBB):
The LI-COR Odyssey CLx is a near-infrared imaging system that produces consistent and reproducible digital images. Both strong and faint bands are clearly visible within the same image, and images present with appreciable sensitivity and no image saturation. Multiplexing with two fluorescent colors is possible. Near-infrared fluorescence (700nm or 800nm) produces consistent signals that are not negatively affected by timing, and the system yields 6 logs of linear dynamic range. Scan up to 9 mini-blots, multiple 96 well or 384 well plates, or 30 slides simultaneously. Applications include western blotting, protein & gel shift assays, as well as tissue section imaging.
Cell Analysis & Automated Cell Culture
Offering particle detection with an overall cell size range of 0.4 um to 1,200 um, the Multisizer offers great flexibility for most routine applications. With aperture size ranges from 20 uM to 2 mm, analyze complex samples with wide particle size distribution. In addition, the instrument exhibits accurate particle density even in non-aqueous solvent systems.
AMBR 15 Microbioreactor for Automated Cell Culture:
The AMBR 15 Microbioreactor mimics the characteristics of classical bioreactors at microscale (10-15ml max volume), by using cost effective, disposable micro bioreactors, controlled by an automated workstation with liquid handler. The system enables the rapid evaluation of stir speed, media type, pH, & DO for increased productivity in cell line development with significant savings on materials and labor.
With 24 disposable bioreactors, users can perform parallel processing and evaluation of multiple experiments in a well controlled, automated benchtop system that is housed within a Biosafety Cabinet. Applications include cell line selection, process optimization, media development, media feed strategies, micro-carrier experiments, and transient transfection.
